I know how to increase demand and all that but getting the major industry businesses is a thing i have never mastered. id prefer to have a factory that employs 150 rather then ten small mixing plants that employ 15 each.

I don't know if there is a way to have influence on this. I just post this, because I thought of something else: on Simtropolis you can find a modd that quadruples the number of jobs provided by the industrial buildings. This means that you don't have to zone so much industry. Visually nothing will change. It is called, you guessed it, the Industry Quadrupler.
I suppose you could download some bigger Industrial buildings so making the chance of getting factories etc more higher than before. This is only a suggestion
The only time Ive seen a large factory is when i zoned large density right by a dirty factory. the rest of the time the medium density seems to yield the same amount of jobs as the dense industrial. It's all about having high-tech in you big city and dirty manufacturing supporting it in neighboring cities IMO. You're going to have to dedicate some large amount of land to generate enough industrial jobs, just a part of the game.
Also try making a city with lots of farms, enough to get the state fair. you have to dedicate a city to your region for this, but i think it's worth it. you wont get alot of industrial jobs, but you'll curb the agriculture demand some.
As far as generating industrial demand, seaports and train tracks with freight stations can increase industrial demand as well as a well-educated workforce and low industrial taxes.
